2: Can you give the chocolate to a male npc (such as Miroku) if your character is also male?

You can't give chocolate until Feb 14th.
Female partners already have set presents that you can receive and I don't see CS making a gender specific event, so yes, there probably will be some yaoi action going on.

Get three Sweets Ingrediments from Shop Owner Mitsuba, and give them to Hasekura Tsunenaga in exchange for the Valentines Day Chocolate. (These are the two new male NPCs in Onigashima)

Ah sorry, I'm not aware of a sugar item for this event o_o I haven't done any grinding lately.

Should be all girls. JP website states to talk to everyone to get their presents. I also assume that talking to them is the only condition to get their presents.
